چکیده

www.aacrjournals.org ABSTRACT Mutations in KRAS drive the oncogenic phenotype in a variety of tumors of epithelial origin. The NF-κB transcription factor pathway is important for oncogenic RAS to transform cells and to drive tumorigenesis in animal models. Recently, TGF-β–activated kinase 1 (TAK1), an upstream regulator of IκB kinase (IKK), which controls canonical NF-κB signaling, was shown to be important for chemoresistance in pancreatic cancer and for regulating KRAS-mutant colorectal cancer cell growth and survival. Here, we show that mutant KRAS upregulates glycogen synthase kinase 3α (GSK-3α), leading to its interaction with TAK1 to stabilize the TAK1–TAB complex to promote IKK activity. In addition, GSK-3α is required for promoting critical noncanonical NF-κB signaling in pancreatic cancer cells. Pharmacologic inhibition of GSK-3 suppresses growth of human pancreatic tumor explants, consistent with the loss of expression of oncogenic genes such as c-myc and TERT . These data identify GSK-3α as a key downstream effector of oncogenic KRAS via its ability to coordinately regulate distinct NF-κB signaling pathways.

GSK-3β Governs Inflammation-Induced NFATc2 Signaling Hubs to Promote Pancreatic Cancer Progression.

We aimed to investigate the mechanistic, functional, and therapeutic role of glycogen synthase kinase 3β (GSK-3β) in the regulation and activation of the proinflammatory oncogenic transcription factor nuclear factor of activated T cells (NFATc2) in pancreatic cancer. IHC, qPCR, immunoblotting, immunofluorescence microscopy, and proliferation assays were used to analyze mouse and human tissues a...

Human Cancer Biology Activation of NF-kB Is a Novel Target of KRAS-Induced Endometrial Carcinogenesis

Purpose: Although the KRAS mutation is one of critical genetic alterations in endometrial carcinogenesis, the downstream targets are not known. Experimental Design: In this study, we investigated the molecular targets of KRAS signals, using tumorigenic cells with oncogenic KRASmutation established from telomerase reverse transcriptase (TERT)immortalized endometrial epithelial cells. Results: We...

Cancer Biology Select

Signal transduction pathways commonly go awry in cancer. Recent advances in understanding how these pathways contribute to tumor formation are the focus of this issue's Cancer Biology Select. New studies use RNAi-based screens and powerful mouse models to pull out critical factors essential for the survival of lung cancer cells. Other discoveries include a chemical-genomics approach to screen f...

Purpose:Death-associated protein kinase 1 (DAPK1), a tumor suppressor, is a rate-limiting effector in an endoplasmic reticulum(ER) stress-dependent apoptotic pathway. Its expression is epigenetically suppressed in several tumors. Amechanistic basis for epigenetic/transcriptional repressionofDAPK1was investigated in certain forms of acute myeloid leukemia (AML) with poor prognosis, which lacked ...

Overview on how oncogenic Kras promotes pancreatic carcinogenesis by inducing low intracellular ROS levels

Pancreatic ductal adenocarcinoma (PDAC) is a devastating disease without clearly known disease causes. Recent epidemiological and animal studies suggest that the supplementation of dietary antioxidants (e.g., vitamins C and E) decreases cancer risk, implying that increased reactive oxygen species (ROS) may play a role in pancreatic carcinogenesis. However, oncogenic Kras mutations (e.g., Kras(G...
